Hidrolisis Enzimatik Ampas Tebu (Bagasse) Memanfaatkan Enzim Selulase dari Mikrofungi Trichoderma reseei dan Aspergillus niger Sebagai Katalisator dengan Pretreatment Microwave
Abstract
Ampas tebu (bagasse) adalah salah satu sumber lignoselulosa untuk pembuatan bioetanol. Pada penelitian hidrolisis dilakukan secara enzimatik dengan memanfaatkan enzim selulase dari mikrofungi Trichoderma reseei dan Aspergillus niger. Proses hidrolisis dilakukan dengan memvariasikan perbandingan dari kedua enzim selulase serta pengamatan terhadap waktu hidrolisis. Variasi perbandingan volume enzim selulase antara Trichodermai reesei : Aspergillus niger yaitu 1:0, 0:1, 1:1, 1:2, 2:1, sedangkan untuk waktu pengambilan sampel dilakukan pada jam ke- 6, 12, 18, 24, 30, 36, 42, serta pada jam ke-48. Pengukuran kadar glukosa dilakukan dengan metode DNS (Dinitrosalicylic acid) dengan menggunakan spektrofotometer pada panjang gelombang 540 nm. Hasil penelitian menunjukkan bahwa perlakuan dengan hasil tertinggi diperoleh pada variasi dengan perbandingan 0 T.reesei : 1 A.niger dengan waktu hidolisis selama 12 jam yang menghasilkan glukosa sebesar 47,213 %.
